Here, we investigated the effect of amyloid peptides on chemokine-induced upregulation of adhesion molecules on the microglial cell line N11. We show that amyloid beta-peptides (A beta) are able to inhibit the expression of the adhesion molecules intercellular adhesion molecule-1 (ICAM-1), leukocyte function-associated antigen-1 (LFA-1), and very late antigen-4 (VLA-4) on N11 cells stimulated with the chemokine interferon-gamma-inducible protein-10/CXCL10. The human neuroblastoma cell line SH-SY5Y, having a neuronal phenotype, was not sensitive to A beta. The peptide inhibitory effect on cytokine-induced upregulation of adhesion molecules was not due to the ability of A beta to block cytokine binding to the cell surface, but was found to be mediated through the A beta-receptor-mediated pathway.
